Description
CHAPS (3-[(3-Cholamidopropyl)-dimethylammonio]-1-propanesulfonate) is a zwitterionic detergent derived from the steroidal scaffold of cholic acid, combining mild solubilizing power with exceptional protein compatibility. Its unique balance of hydrophobic and hydrophilic domains preserves the native conformation of membrane proteins during extraction and reconstitution. Unlike ionic detergents, CHAPS minimizes protein denaturation and is ideal for isolating membrane complexes, studying lipid–protein interactions, and stabilizing liposomes. This detergent’s cholic acid-based ring system provides structural rigidity, while the sulfonate headgroup ensures solubility, making CHAPS a benchmark reagent in structural biology, protein crystallization, and membrane biochemistry research.

Application
Used as a non-denaturing biological detergent; CHAPS is a zwitterionic detergent used in the laboratory to solubilize biological macromolecules such as proteins. It is used as a non-denaturing solvent in the process of protein purification and is especially useful in purifying membrane proteins, which are often sparingly soluble or insoluble in aqueous solution due to their native hydrophobicity.
